\n\u570b\u7acb\u7af9\u5317\u9ad8\u7d1a\u4e2d\u5b78\u751f\u7269\u79d1\u5f35\u96c5\u83f1\u8001\u5e2b\/\u570b\u7acb\u81fa\u7063\u5e2b\u7bc4\u5927\u5b78\u751f\u547d\u79d1\u5b78\u7cfb\u674e\u51a0\u7fa4\u52a9\u7406\u6559\u6388\u8cac\u4efb\u7de8\u8f2f<\/span><\/strong><\/p>\n

\u805a\u5408\u9176\u9023\u9396\u53cd\u61c9 (Polymerase chain reaction, PCR) \u662f\u7576\u4ee3\u4e00\u9805\u91cd\u8981\u7684\u751f\u7269\u6280\u8853\uff0c\u53ef\u4ee5\u5c07\u55ae\u4e00\u6216\u5c11\u8a31\u7684DNA\u7247\u6bb5\uff0c\u7d93\u7531\u4e00\u7cfb\u5217\u7684\u6eab\u5ea6\u8b8a\u5316\u8655\u7406\u4f7f\u5176\u4e0d\u65b7\u5730\u8907\u88fd\uff0c\u6700\u5f8c\u5f97\u5230\u5927\u91cf\u4e14\u76f8\u540c\u7684DNA\u7247\u6bb5\u3002\u52a0\u7406\u2027\u83ab\u7406\u65af (Kary B. Mullis) \u662f\u9019\u9805\u6280\u8853\u7684\u767c\u660e\u8005\u3002<\/p>\n

1983\u5e74\u6642\uff0c\u4ed6\u5728\u52a0\u5dde\u7684Cetus\u751f\u7269\u79d1\u6280\u516c\u53f8\u8ca0\u8cac\u77ed\u7247\u6bb5DNA\u7684\u5408\u6210\u3002\u7576\u6642\u8981\u505aDNA\u76f8\u95dc\u7814\u7a76\u6642\uff0c\u5e38\u5e38\u6703\u9762\u81e8\u5230DNA\u6a23\u672c\u6578\u4e0d\u5920\u7684\u554f\u984c\uff0c\u5982\u6b64\u4e00\u4f86\u5c31\u5fc5\u9808\u82b1\u8cbb\u6642\u9593\u91cd\u65b0\u8403\u53d6DNA\uff0c\u751a\u81f3\u53ef\u80fd\u56e0\u751f\u7269\u6a23\u672c\u6578\u4e0d\u8db3\uff0c\u800c\u7121\u6cd5\u7e7c\u7e8c\u5be6\u9a57\u3002<\/p>\n

\u83ab\u7406\u65af\u8868\u793a\uff0c\u5728\u67d0\u500b\u591c\u665a\uff0c\u7576\u4ed6\u958b\u8eca\u884c\u99db\u5728\u592a\u5e73\u6d0b\u6d77\u5cb8\u516c\u8def(Pacific Coast Highway)\u4e0a\u6642\uff0c\u601d\u8003\u8457\u5982\u4f55\u8907\u88fdDNA\uff0c\u7a81\u7136\u9748\u6a5f\u4e00\u52d5\uff1a\u539f\u6709\u7684\u5c11\u8a31DNA\u7247\u6bb5\uff0c\u7d93\u7531\u91cd\u8907\u5347\u6eab\u3001\u964d\u6eab\u7c21\u55ae\u7684\u6b65\u9a5f\uff0c\u5c31\u80fd\u4f7fDNA\u7247\u6bb5\u5f9e\u4e00\u689d\u8b8a\u5169\u689d\uff0c\u5169\u689d\u8b8a\u56db\u689d\uff0c\u56db\u689d\u8b8a\u516b\u689d\uff0c\u4ee5\u6b64\u985e\u63a8\u3002\u5982\u6b64\u4e00\u4f86\uff0c\u53ea\u9700\u4e00\u500b\u4e0b\u5348\u7684\u6642\u9593\uff0c\u5c31\u80fd\u5728\u4e00\u500b\u5c0f\u5c0f\u7684\u8a66\u7ba1\u5167\uff0c\u88fd\u9020\u51fa100\u5104\u500b\u76f8\u4f3c\u7684DNA\u7247\u6bb5\uff01\u4e00\u958b\u59cb\u83ab\u7406\u65af\u8a8d\u70ba\u9019\u9ebc\u7c21\u55ae\u7684\u65b9\u6cd5\u53ef\u80fd\u65e9\u5c31\u6709\u4eba\u60f3\u51fa\u4f86\u4e86\uff0c\u4f46\u56de\u5230\u5be6\u9a57\u5ba4\u67e5\u95b1\u6587\u737b\u4e4b\u5f8c\uff0c\u624d\u767c\u73fe\u4e26\u6c92\u6709\u4eba\u505a\u904e\u985e\u4f3c\u7684\u5be6\u9a57\u3002<\/p>\n

\u96a8\u5f8c\uff0c\u4ed6\u5617\u8a66\u4e00\u9023\u4e32\u7684\u5be6\u9a57\uff0c\u7d42\u65bc\u8b93\u9019\u500b\u8907\u88fdDNA\u7684\u60f3\u6cd5\u6210\u70ba\u53ef\u80fd\uff01\u83ab\u7406\u65af\u5c07\u9019\u9805\u767c\u660e\u767c\u8868\u5728\u8457\u540d\u7684\u300a\u79d1\u5b78\u300b(Science) \u00a0\u671f\u520a\u4e0a\u3002\u8ad6\u6587\u767c\u8868\u5f8c\uff0cPCR\u6280\u8853\u958b\u59cb\u53d7\u5230\u5404\u754c\u7684\u95dc\u6ce8\uff0c\u4e26\u4e14\u7d93\u904e\u4e0d\u65b7\u6539\u9032\uff0c\u5ee3\u6cdb\u61c9\u7528\u5728\u751f\u7269\u79d1\u6280\u8207\u91ab\u5b78\u7814\u7a76\u4e0a\u3002\u800c\u9019\u9805\u7c21\u55ae\u53c8\u5049\u5927\u7684\u767c\u660e\uff0c\u4e5f\u8b93\u83ab\u7406\u65af\u7372\u98121993\u5e74\u7684\u8afe\u8c9d\u723e\u5316\u5b78\u734e\u3002<\/p>\n

PCR\u653e\u5927DNA\u7247\u6bb5\u7684\u65b9\u6cd5\u5982\u4e0b\uff1a\u9996\u5148\u5229\u7528\u52a0\u71b1\u7684\u65b9\u5f0f (\uff1e90\u2103) \u5c07\u96d9\u80a1DNA\u5206\u958b\uff0c\u7576\u4f5c\u6a21\u7248DNA\uff08template DNA\uff0c\u9700\u8981\u88ab\u653e\u5927\u7684DNA\u7247\u6bb5\uff09\u3002<\/p>\n

\u52a0\u5165\u80fd\u8207\u6a21\u7248DNA\u5169\u7aef\u4e92\u88dc\u7684\u5be1\u6838\u82f7\u9178\u7247\u6bb5\uff0c\u7a31\u70ba\u300c\u5f15\u5b50 (primer)\u300d\u3002<\/p>\n
