dehydrogenase complex\uff09\u8f49\u63db\u70ba\u4e59\u91af\u8f14\u9176 A\uff08acetyl-CoA\uff09\uff0c\u518d\u9001\u5165\u6ab8\u6aac\u9178\u5faa\u74b0\uff08Citric acid cycle\uff09\u5b8c\u5168\u6c27\u5316\u3002<!--more--><\/p>\n<p>\u7f3a\u6c27\u7684\u6642\u5019\u56e0\u70ba\u7c92\u7dda\u9ad4\u7121\u6cd5\u4f5c\u7528\uff0c\u53ea\u5269\u4e0b\u91a3\u89e3\u4f5c\u7528\u7368\u6311\u5927\u6a11\uff1b\u4f46\u662f\u91a3\u89e3\u4f5c\u7528\u9700\u8981\u4e0d\u65b7\u7684\u6709\u83f8\u9e7c\u91af\u80fa\u817a\u560c\u5464\u4e8c\u6838\u82f7\u9178\uff08Nicotinamide adenine dinucleotide\uff0c\u4ee5\u4e0b\u7c21\u7a31 NAD<sup>+<\/sup>\uff09\u5c07\u5b83\u7522\u751f\u7684\u96fb\u5b50\u5e36\u8d70\u3002\u539f\u672c\u9019\u500b\u5de5\u4f5c\u662f\u4ea4\u7d66\u7c92\u7dda\u9ad4\u8655\u7406\u7684\uff0c\u4f46\u7f3a\u6c27\u6642\u7c92\u7dda\u9ad4\u7121\u6cd5\u5de5\u4f5c\uff0c\u65bc\u662f\u96fb\u5b50\u4fbf\u4ee5 NADH \u7684\u578b\u614b\u7d2f\u7a4d\uff0c\u9020\u6210\u91a3\u89e3\u4f5c\u7528\u56e0\u70ba\u7f3a\u5c11 NAD<sup>+\u00a0<\/sup>\u800c\u7121\u6cd5\u5de5\u4f5c\u3002<\/p>\n<p>\u9019\u6642\u5019\u6211\u5011\u7684\u7d30\u80de\u6703\u5148\u9032\u884c\u767c\u9175\u4f5c\u7528\uff08fermentation\uff09\uff0c\u6d88\u8017\u6389\u4e00\u90e8\u4efd\u7684 NADH\uff0c\u4f7f\u91a3\u89e3\u4f5c\u7528\u53ef\u4ee5\u7e7c\u7e8c\u904b\u884c\uff1b\u4f46\u662f\u767c\u9175\u4f5c\u7528\u6703\u7522\u751f\u4e73\u9178\uff08lactic acid\uff09\uff0c\u4e73\u9178\u5e36\u6709\u8f15\u5fae\u7684\u6bd2\u6027\uff0c\u4e0d\u80fd\u5132\u5b58\u5728\u9ad4\u5167\uff0c\u5fc5\u9808\u8981\u76e1\u5feb\u4ee3\u8b1d\u6389\uff0c\u5426\u5247\u5c31\u6703\u767c\u751f\u4e73\u9178\u4e2d\u6bd2\uff08lactic acidosis\uff09\uff1b\u56e0\u6b64\uff0c\u810a\u690e\u52d5\u7269\u4e0d\u80fd\u5728\u7f3a\u6c27\u7684\u72c0\u6cc1\u4e0b\u300c\u6490\u300d\u5f88\u4e45\u3002<\/p>\n<p>\u90a3\u9ebc\u9019\u4e9b\u53b2\u5bb3\u7684\u9b5a\uff08\u5982\u9ed1\u9bfd\u00a0<em>Carassius carassius<\/em>\u6216\u91d1\u9b5a\u00a0<em>Carassius auratus<\/em>\uff09\u662f\u600e\u9ebc\u6490\u597d\u5e7e\u500b\u6708\u5462\uff1f<\/p>\n<p>\u904e\u53bb\u7684\u7814\u7a76\u767c\u73fe\uff0c\u7260\u5011\u7adf\u7136\u53ef\u4ee5\u628a\u4e73\u9178\u8f49\u70ba\u9152\u7cbe\uff0c\u7136\u5f8c\u518d\u7d93\u7531\u9c13\u6392\u5230\u6c34\u4e2d\uff01\u4e4b\u524d\u7684\u7814\u7a76\u5718\u968a\u8a8d\u70ba\uff0c\u53ef\u80fd\u662f PDH \u628a\u4e19\u916e\u9178\u8f49\u70ba\u4e59\u919b\uff0c\u518d\u7531\u4e00\u500b\u7279\u5225\u7684\u9152\u7cbe\u53bb\u6c2b\u9176\uff08ADH\uff0calcohol dehydrogenase\uff09\u5c07\u4e59\u919b\u8f49\u70ba\u9152\u7cbe\u3002<\/p>\n<p>PDH \u662f\u771f\u6838\u751f\u7269\u4e2d\u6700\u5927\u7684\u86cb\u767d\u8cea\u8907\u5408\u9ad4\uff08protein complex\uff09\uff0c\u7531\u4e09\u500b\u9175\u7d20 E1, E2, E3 \u69cb\u6210\u3002\u5176\u4e2d\u6700\u91cd\u8981\u7684\u662f E1\uff0c\u7531\u5169\u500b\u03b1\u8207\u5169\u500b\u03b2\u6b21\u55ae\u5143\uff08subunit\uff09\u69cb\u6210\uff0c\u5176\u529f\u80fd\u70ba\u5c07\u4e19\u916e\u9178\u5148\u812b\u78b3\u3001\u9084\u539f\u5f8c\u518d\u5c07\u4e59\u91af\u57fa\uff08acetyl group\uff09\u8f49\u5230 E2\uff1b\u5982\u679c\u91d1\u9b5a\u7684 PDH \u4e0d\u518d\u5c07\u4e19\u916e\u9178\u812b\u78b3\u3001\u9084\u539f\uff0c\u800c\u662f\u5c07\u4e19\u916e\u9178\u8f49\u70ba\u4e59\u919b\uff0c\u662f\u5426\u610f\u5473\u8457 E1 \u7522\u751f\u4e86\u8b8a\u5316\u5462\uff1f<\/p>\n<p>\u70ba\u4e86\u89e3\u958b\u9019\u500b\u8b0e\u984c\uff0c\u632a\u5a01\u7684\u79d1\u5b78\u5bb6\u7d44\u6210\u7684\u7814\u7a76\u5718\u968a\u6bd4\u8f03\u4e86\u9bc9\u79d1\u9b5a\u985e\uff08\u9bc9\u9b5a\u3001\u9ed1\u9bfd\u3001\u91d1\u9b5a\uff09\u7684 PDH \u8207 ADH \u7684\u4fe1\u606f\u6838\u91a3\u6838\u9178\uff08mRNA\uff0cmessenger RNA\uff09\u7684\u5e8f\u5217\uff0c\u4ee5\u53ca\u9019\u4e9b\u57fa\u56e0\u5728\u6b63\u5e38\u72c0\u6cc1\u3001\u7f3a\u6c27\u4e00\u5929\u3001\u7f3a\u6c27\u4e03\u5929\u3001\u7f3a\u6c27\u4e03\u5929\u5f8c\u518d\u653e\u56de\u6b63\u5e38\u72c0\u6cc1\u516d\u5929\u5728\u4e0d\u540c\u7d44\u7e54\u88e1\u7684\u8868\u73fe\u72c0\u6cc1\u3002<\/p>\n<p>\u7d50\u679c\u767c\u73fe\uff0c\u5728\u9bc9\u9b5a\u7684\u4e0d\u540c\u7d44\u7e54\u88e1\uff0c\u4e0d\u8ad6\u4ec0\u9ebc\u72c0\u6cc1\uff0c\u7260\u7684 E1, E2, E3 \u7684\u8868\u73fe\u91cf\u90fd\u5dee\u4e0d\u591a\uff1b\u4f46\u662f\u5728\u9ed1\u9bfd\u7684\u9aa8\u9abc\u808c\u4e2d\uff0c\u5176 E1\u03b1 \u8207 E1\u03b2 \u7684 mRNA \u6574\u9ad4\u8868\u73fe\u91cf\u662f\u5fc3\u3001\u809d\u3001\u8166\u7684\u5341\u500d\u5230\u4e00\u767e\u500d\u3002\u86cb\u767d\u8cea\u7684\u8868\u73fe\u91cf\u4e5f\u547c\u61c9 mRNA \u7684\u91cf\u3002<\/p>\n<p>\u9019\u9ebc\u591a E1\u03b1 \u8207 E1\u03b2\uff0c\u662f\u4e0d\u662f\u90fd\u662f\u540c\u4e00\u500b\u57fa\u56e0\u7684\u7522\u7269\u5462\uff1f\u904e\u53bb\u5df2\u77e5\u9b5a\u985e\u5728\u4e00\u81f3\u4e8c\u5104\u5e74\u524d\u66fe\u767c\u751f\u904e\u4e00\u6b21\u57fa\u56e0\u9ad4\u91cd\u8907\uff0c\u9020\u6210 E1\u03b1 \u6709\u5169\u500b\u57fa\u56e0\uff08\u4ee5E1\u03b1<sub>1<\/sub>\u3001E1\u03b1<sub>2<\/sub>\u8868\u793a\uff09\uff0cE2 \u8207 E3 \u4e5f\u5404\u6709\u5169\u500b\uff08\u4ee5 E2a\u3001E2b\u3001E3a\u3001E3b \u8868\u793a\uff09\uff1b\u800c E1\u03b2 \u53ea\u6709\u4e00\u500b\u3002\u7814\u7a76\u5718\u968a\u5206\u6790\u5e8f\u5217\u7684\u7d50\u679c\u767c\u73fe\uff0c\u5728\u9ed1\u9bfd\u8207\u91d1\u9b5a\u7684\u9aa8\u9abc\u808c\u4e2d\uff0c\u9664\u4e86\u672c\u4f86\u7684\u5169\u500b E1\u03b1 \u8207\u4e00\u500b E1\u03b2 \u4ee5\u5916\uff0c\u9084\u591a\u4e86\u4e00\u500b E1\u03b1 \u8207 E1\u03b2\uff08\u4ee5 E1\u03b1<sub>3\u00a0<\/sub>\u8207 \u7684\u6b21\u55ae\u5143\u7684\u8868\u73fe\u91cf\u90fd\u5f88\u4f4e\u3002<\/p>\n<p>\u591a\u51fa\u4f86\u7684 E1\u03b1 \u8207 E1\u03b2 \u662f\u54ea\u88e1\u4f86\u7684\u5462\uff1f\u7814\u7a76\u5718\u968a\u8a8d\u70ba\uff0c\u9019\u5169\u500b\u65b0\u7684\u57fa\u56e0\u61c9\u8a72\u4f86\u81ea\u65bc 820 \u842c\u5e74\u524d\u7684\u9bc9\u79d1\u9b5a\u985e\u57fa\u56e0\u9ad4\u91cd\u8907\u4e8b\u4ef6\u3002\u5f9e\u6591\u99ac\u9b5a\uff08<em>Danio rerio<\/em>\uff09\u7684\u57fa\u56e0\u9ad4\u4e5f\u5370\u8b49\u4e86\u9019\u9ede\u3002\u6591\u99ac\u9b5a\u5728\u9bc9\u79d1\u9b5a\u985e\u57fa\u56e0\u9ad4\u91cd\u8907\u767c\u751f\u4e4b\u524d\uff0c\u5c31\u8207\u7260\u5011\u5206\u9053\u63da\u9463\uff1b\u5206\u6790\u7260\u7684\u57fa\u56e0\u9ad4\u4e5f\u53ea\u80fd\u770b\u5230\u5169\u500b E1\u03b1 \u8207\u4e00\u500b E1\u03b2\u3002<\/p>\n<p>\u4ee5\u4e0a\u9019\u4e9b\u767c\u73fe\u8b93\u7814\u7a76\u5718\u968a\u8a8d\u70ba\uff1a\u91d1\u9b5a\u8207\u9ed1\u9bfd\u5c07\u4e19\u916e\u9178\u8f49\u5316\u70ba\u4e59\u919b\u7684\u80fd\u529b\uff0c\u53ef\u80fd\u5c31\u662f\u4f86\u81ea\u65bc\u9019\u5169\u500b\u591a\u51fa\u4f86\u7684\u57fa\u56e0\uff1b\u4e5f\u5c31\u662f\u8aaa\uff0cE1\u03b1<sub>3\u00a0<\/sub>\u8207 E1\u03b2<sub>2\u00a0<\/sub>\u7d44\u6210\u4e00\u500b\u7279\u5225\u7684 PDH-E1\uff0c\u5c07\u4e19\u916e\u9178\u8f49\u5316\u70ba\u4e59\u919b\u3002<\/p>\n<p>\u662f\u5426\u9019\u7279\u5225\u7684 PDH-E1\uff0c\u771f\u7684\u53ef\u4ee5\u7368\u7acb\u4f5c\u6230\u5462\uff1f\u9996\u5148\u7814\u7a76\u5718\u968a\u5206\u6790\u4e86\u9019\u5169\u500bE1\u6b21\u55ae\u5143\u7684\u8868\u73fe\u91cf\uff0c\u767c\u73fe\u5728\u5fc3\u3001\u809d\u3001\u8166\u4e2d\u7684 E1:E2:E3 \u7684\u6bd4\u4f8b\u90fd\u5dee\u4e0d\u591a\uff1b\u4f46\u662f\u5728\u7d05\u808c\u8089\u8207\u767d\u808c\u8089\u4e2d\u4e0d\u8ad6\u662f E1:E2 \u6216 E1:E3 \u90fd\u51fa\u73fe\u516d\u5341\u500d\u7684\u5dee\u8ddd\u3002<\/p>\n<p>\u9019\u9ebc\u591a\u7684 E1\uff0c\u7576\u7136\u6709\u53ef\u80fd\u6709\u4e00\u4e9bE1\u662f\u7368\u7acb\u4f5c\u6230\u8005\uff1b\u4f46\u662f\u7814\u7a76\u5718\u968a\u63a5\u8457\u5206\u6790 E1\u03b1<sub>3\u00a0<\/sub>\u8207 E1\u03b2<sub>2\u00a0<\/sub>\u7684\u5e8f\u5217\uff0c\u767c\u73fe E1\u03b2<sub>2\u00a0<\/sub>\u7684\u57fa\u56e0\u6709\u500b\u7a81\u8b8a\uff0c\u4f7f\u5b83\u7684\u7b2c 319 \u865f\u6c28\u57fa\u9178\u7531\u5e36\u96fb\u7684\u5929\u51ac\u80fa\u9178\uff08aspartic acid\uff0cD\uff09\u8b8a\u6210\u4e0d\u5e36\u96fb\u7684\u5929\u51ac\u9170\u80fa\uff08asparagine\uff0cN\uff09\u3002\u904e\u53bb\u5728\u4eba\u985e\u7684 E1\u03b2 \u4e2d\u66fe\u767c\u73fe\u985e\u4f3c\u7684\u7a81\u8b8a\uff0c\u9020\u6210 E1 \u8207 E2 \u5206\u96e2\uff0c\u5f71\u97ff PDH \u7684\u529f\u80fd\u3002\u9019\u6a23\u7684\u7a81\u8b8a\u5c0d\u4e00\u822c\u7684 PDH \u662f\u6709\u5bb3\u7684\uff0c\u4f46\u5c0d\u65bc\u91d1\u9b5a\u8207\u9ed1\u9bfd\u7684 PDH-E1 \u4f86\u8aaa\uff0c\u537b\u662f\u6b63\u4e2d\u4e0b\u61f7\uff01\u8ddf\u5176\u4ed6 PDH \u7684 E1 \u985e\u4f3c\u7684\u662f\uff0c\u9019\u500b PDH-E1 \u4e5f\u662f\u900f\u904e\u8abf\u7bc0 \u03b1 \u6b21\u55ae\u5143\u7684\u6d3b\u6027\u4f86\u8abf\u7bc0\u6574\u500b\u9175\u7d20\u7684\u6d3b\u6027\u3002\u5728\u7f3a\u6c27\u6642\uff0c\u5b83\u7684 E1\u03b1 \u6703\u88ab\u53bb\u78f7\u9178\u5316\u800c\u6d3b\u5316\uff1b\u7576\u6c27\u6c23\u91cf\u6062\u5fa9\u6b63\u5e38\u5f8c\uff0c\u5b83\u53c8\u6703\u88ab\u78f7\u9178\u5316\u800c\u5931\u53bb\u6d3b\u6027\u4e86\u3002<\/p>\n<p>\u6240\u4ee5\uff0c\u5728\u91d1\u9b5a\u8207\u9ed1\u9bfd\u4e2d\u591a\u51fa\u4f86\u7684\u9019\u500b PDH-E1\uff0c\u5b83\u5e36\u6709 D319N \u7a81\u8b8a\u4f7f\u5b83\u7121\u6cd5\u8207 E2 \u7d50\u5408\uff1b\u4f46\u9019\u9ebc\u4e00\u4f86\u537b\u6b63\u597d\u8b93\u5b83\u53ef\u4ee5\u767c\u63ee\u6240\u9577\uff0c\u5c07\u4e19\u916e\u9178\u8f49\u63db\u70ba\u4e59\u919b\u3002<\/p>\n<p>\u5149\u662f\u8f49\u63db\u70ba\u4e59\u919b\u9084\u662f\u4e0d\u5920\uff0c\u5982\u679c\u4e59\u919b\u4e0d\u80fd\u88ab\u8f49\u63db\u70ba\u9152\u7cbe\uff0c\u90a3\u9ebc\u4e5f\u53ea\u662f\u6bd2\u5bb3\u81ea\u5df1\u800c\u5df2\u3002\u9019\u5c31\u9700\u8981\u4e00\u500b\u80fd\u5920\u628a\u4e59\u919b\u8f49\u63db\u70ba\u9152\u7cbe\u7684\u4e59\u9187\u53bb\u6c2b\u9176\uff08ADH\uff09\u3002\u5728\u6591\u99ac\u9b5a\u4e2d\u78ba\u5be6\u6709\u9019\u9ebc\u4e00\u500bADH\uff0c\u7a31\u70baADH8a\uff1b\u4f46\u5b83\u53ea\u8868\u73fe\u5728\u809d\u81df\u88e1\u3002\u662f\u5426\u91d1\u9b5a\u8207\u9ed1\u9bfd\u7684\u57fa\u56e0\u9ad4\u4e2d\u4e5f\u6709\u8868\u73fe\u5728\u808c\u8089\u7684ADH8a\u5462\uff1f<\/p>\n<p>\u7814\u7a76\u5718\u968a\u767c\u73fe\uff0c\u5728\u91d1\u9b5a\u8207\u9ed1\u9bfd\u7684\u57fa\u56e0\u9ad4\u4e2d\uff0c\u5b58\u5728\u8457\u4e09\u500b
