June's et al. Science 337,816-821;2012)\uff0c\u65bc2012 \u5e745\u670825\u65e5\u958b\u59cb\u4ed6\u5011\u7684\u5c08\u5229\u7533\u8acb\u3002\u53e6\u4e00\u5718\u968a\u5247\u662f\u7531\u5f35\u92d2\u6240\u5e36\u9818\u7684\u9ebb\u7701\u7406\u5de5\u5b78\u9662\u548c\u54c8\u4f5b\u5927\u5b78\u7684Broad \u7814\u7a76\u9662\u5718\u968a\uff0c\u4ed6\u5011\u65bc2013\u5e74\u767c\u8868\u5c07 CRISPR-Cas9\u61c9\u7528\u5728\u54fa\u4e73\u985e\u52d5\u7269\u7d30\u80de\u7684\u8ad6\u6587\uff08L. Cong et al. science 339,819-823;2013)\u3002\u4e26\u65bc2012\u5e7412\u670812\u65e5\u63d0\u51fa\u5c08\u5229\u7533\u8acb\u3002<\/p>\n<p>\u96d6\u7136\u67cf\u514b\u840a\u5927\u5b78\u5718\u968a\u5148\u63d0\u51fa\u7533\u8acb\uff0c\u4f46\u662f Broad \u5718\u968a\u5c07\u4ed6\u5011\u7684\u5c08\u5229\u7533\u8acb\u4ee5\u52a0\u6025\u5be9\u67e5\u65b9\u6848\u63d0\u4ea4\u8655\u7406\uff0c\u4e26\u65bc2014\u5e74\u56db\u6708\u7372\u5f97\u5c08\u5229\u3002\u67cf\u514b\u840a\u5927\u5b78\u5718\u968a\u65bc\u662f\u63d0\u51fa\u5c08\u5229\u885d\u7a81\u8a34\u8a1f\uff0c\u5c0d\u6297 Broad \u7684\u539f\u5c08\u5229\u4ee5\u53ca\u5341\u4e00\u9805\u76f8\u95dc\u7684\u5c08\u5229\uff0c\u7f8e\u570b\u5c08\u5229\u548c\u5546\u6a19\u5c40\u65bc2016\u5e741\u670811\u65e5\u6b63\u5f0f\u53d7\u7406\u67cf\u514b\u840a\u5927\u5b78\u5718\u968a\u7684\u8981\u6c42\u3002<\/p>\n<p>\u7f8e\u570b\u5c08\u5229\u7684\u7533\u8acb\u539f\u672c\u662f\u7d66\u7b2c\u4e00\u500b\u767c\u660e\u8005\uff08first-to-invent)\uff0c\u4f46\u662f\u57282013\u5e74\uff0c\u66f4\u6539\u70ba\u4ee5\u7b2c\u4e00\u500b\u7533\u8acb\u8005\u70ba\u5148\uff08first-to-file system)\u4e0d\u4e00\u6a23\u7684\u7cfb\u7d71\u3002\u4f46\u662f\u5e7e\u500b CRISPR-Cas9 \u7684\u95dc\u9375\u6027\u5c08\u5229\u662f\u5728\u6539\u5236\u4e4b\u524d\u5c31\u63d0\u51fa\uff0c\u56e0\u6b64\u9020\u6210\u5c08\u5229\u885d\u7a81\u8a34\u8a1f\u3002<\/p>\n<p>\u7f8e\u570b\u5c08\u5229\u548c\u5546\u6a19\u5c40\u6703\u6536\u96c6\u96d9\u65b9\u6240\u63d0\u4f9b\u7684\u8b49\u64da\uff0c\u4f86\u6c7a\u5b9a\u54ea\u500b\u5718\u968a\u5148\u5c07CRISPR-Cas9\u61c9\u7528\u5728\u57fa\u56e0\u7de8\u8f2f\u4e0a\u3002\u8655\u7406\u5c08\u5229\u885d\u7a81\u8a34\u8a1f\u53ef\u80fd\u6709\u76f8\u7576\u7684\u6280\u8853\u6027\uff0c\u751a\u81f3\u8981\u8003\u616e\u662f\u54ea\u4e00\u5718\u968a\u5148\u6709\u9019\u6a23\u7684\u60f3\u6cd5\u3002\u65e9\u671f\u8a31\u591a\u516c\u53f8\u751a\u81f3\u4fdd\u5b58\u4e26\u516c\u8b49\u8655\u7406\u5be6\u9a57\u8a18\u9304\u672c\uff0c\u4ee5\u5099\u7533\u8acb\u5c08\u5229\u6709\u722d\u8b70\u6642\u53ef\u63d0\u4f5c\u8b49\u64da\uff0c\u4f46\u662f\u4e00\u822c\u5b78\u8853\u754c\u7684\u5be6\u9a57\u5ba4\u4e26\u6ca1\u6709\u505a\u5230\u9019\u6a23\u7684\u7a0b\u5ea6\uff0c\u8a34\u8a1f\u904e\u7a0b\u53ef\u80fd\u5e7e\u500b\u6708\u751a\u81f3\u5e7e\u5e74\uff0c\u800c\u8f38\u7684\u4e00\u65b9\u53ef\u80fd\u6703\u518d\u63d0\u51fa\u7570\u8b70\uff0c\u6240\u4ee5\u6574\u500b\u904e\u7a0b\u53ef\u80fd\u5ef6\u5b95\u66f4\u4e45\u3002\u6b64\u5916\uff0c\u53ef\u80fd\u6703\u51fa\u73fe\u66f4\u591a\u95dc\u65bc CRISPR-Cas9\u7684\u5c08\u5229\u885d\u7a81\u8a34\u8a1f\uff0c\u91dd\u5c0d\u540c\u4e00\u5c08\u5229\u6709\u591a\u500b\u8a34\u8a1f\u7684\u4f8b\u5b50\u6bd4\u8f03\u5c11\u898b\uff0c\u4f46\u662f\u6709\u53ef\u80fd\u6703\u51fa\u73fe\u3002<\/p>\n<p>\u90a3\u5728\u6b50\u6d32 CRISPR-Cas9 \u7684\u5c08\u5229\u6709\u6bd4\u8f03\u6e05\u695a\u55ce\uff1f\u7b54\u6848\u662f\u5426\u5b9a\u7684\u3002Broad \u5718\u968a\u540c\u6a23\u4e5f\u5728\u6b50\u6d32\u63d0\u51fa\u5c08\u5229\u7533\u8acb\uff0c\u4e26\u4e14\u5df2\u7d93\u7372\u5f97\u6578\u500b\u5c08\u5229\uff0c\u4f46\u662f\u67cf\u514b\u840a\u5718\u968a\u7684\u55ae\u4e00\u5c08\u5229\u7533\u8acb\u4ecd\u5728\u5be9\u67e5\u968e\u6bb5\u3002\u5728\u6b50\u6d32\u96d6\u7136\u6c92\u6709\u885d\u7a81\u8a34\u8a1f\u7a0b\u5e8f\uff0c\u4f46\u662f\u5916\u754c\u53ef\u4ee5\u63d0\u51fa\u7570\u8b70\uff0c\u76ee\u524d\u5df2\u7d93\u6709\u4e5d\u500b\u5718\u9ad4\u5c0d Broad \u5718\u968a\u6240\u6301\u6709\u7684\u7b2c\u4e00\u500b CRISPR-Cas9 \u5c08\u5229\u63d0\u51fa\u7570\u8b70\uff0c\u555f\u52d5\u53cd\u5c0d\u7a0b\u5e8f\uff0c\u53ef\u80fd\u6703\u6301\u7e8c\u591a\u5e74\u624d\u80fd\u89e3\u6c7a\u3002\u4e00\u65e6\u6c7a\u8b70\u51fa\u4f86\uff0c\u53c3\u8207\u8005\u53ef\u4ee5\u518d\u4e0a\u8a34\uff0c\u5982\u6b64\u53ef\u80fd\u5f97\u518d\u82b1\u53e6\u5916\u56db\u5230\u4e94\u5e74\u7684\u6642\u9593\u3002\u6240\u4ee5\u5728\u6b50\u6d32\u4e5f\u662f\u8981\u518d\u5e7e\u5e74\u7684\u6642\u9593\uff0c CRISPR-Cas9 \u7684\u5c08\u5229\u6b78\u5c6c\u624d\u6703\u660e\u6717\u3002<\/p>\n<p>\u96d6\u7136\u76ee\u524d\u5c1a\u672a\u77e5\u9053 CRISPR-Cas9 \u57fa\u56e0\u7de8\u8f2f\u6280\u8853\u7684\u5c08\u5229\u80fd\u70ba\u5be6\u9a57\u5718\u968a\u5e36\u4f86\u591a\u5c11\u5546\u696d\u5229\u76ca\uff0c\u4f46\u662f\u5404\u500b\u7814\u7a76\u5718\u968a\u9084\u662f\u7a4d\u6975\u722d\u53d6\u5c08\u5229\u6b0a\uff0c\u4ee5\u4fdd\u969c\u672a\u4f86\u4ee5\u6b64\u6280\u8853\u767c\u5c55\u51fa\u4f86\u7684\u9f90\u5927\u7d93\u6fdf\u5229\u76ca\u3002<\/p>\n<p>&nbsp;<\/p>\n<p><strong>\u53c3\u8003\u6587\u737b\uff1a<\/strong><br \/>\n1.\u00a0Nature 529, 265 (21 January 2016) doi:10.1038\/nature.2015.17961<br \/>\n2.
