Planck Institute)\u7684\u7814\u7a76\u5718\u968a\uff0c\u767c\u5c55\u51fa\u4e86\u4ee5\u5e72\u64feRNA(siRNA)\u505a\u70ba\u6bba\u87f2\u5291\u7684\u65b9\u6cd5\uff0c\u5e0c\u671b\u53ef\u4ee5\u767c\u5c55\u51fa\u5c08\u9580\u5c0d\u4ed8\u4e00\u7a2e\u5bb3\u87f2\u7684\u6bba\u87f2\u5291\u3002<\/p>\n<p>siRNA\u7684\u6280\u8853\u4e26\u4e0d\u662f\u4ec0\u9ebc\u65b0\u82b1\u6a23\uff1b\u7c21\u55ae\u4f86\u8aaa\uff0c\u5c31\u662f\u8981\u8b93\u4e00\u6bb5\u96d9\u80a1\u7684RNA\u5728\u76ee\u6a19\u7269\u7a2e\u9ad4\u5167\u51fa\u73fe\u3002\u7531\u65bc\u9ad8\u7b49\u751f\u7269\u7d30\u80de\u5167\u901a\u5e38\u4e0d\u6703\u51fa\u73fe\u96d9\u80a1RNA\uff0c\u9664\u975e\u53d7\u5230RNA\u75c5\u6bd2\u611f\u67d3\uff01\u56e0\u6b64\uff0c\u4e00\u65e6\u96d9\u80a1RNA\u51fa\u73fe\u4ee5\u5f8c\uff0c\u5f88\u5feb\u4fbf\u6703\u88ab\u8fa8\u8a8d\u51fa\u4f86\uff0c\u4e26\u4ee5Dicer\u86cb\u767d\u8cea\u8907\u5408\u9ad4\u4fee\u6574\u5f8c\uff0c\u9078\u53d6\u5927\u7d0421\u500b\u6838\u82f7\u9178\u9577\u77ed\u7684\u7247\u6bb5\uff0c\u505a\u70ba\u7d30\u80de\u7684\u300c\u75c5\u6bd2\u624b\u518a\u300d\u3002\u4e0b\u6b21\u6709\u4efb\u4f55RNA\u51fa\u73fe\u5728\u7d30\u80de\u88e1\uff0cDicer\u4fbf\u6703\u62ff\u51fa\u9019\u672c\u300c\u75c5\u6bd2\u624b\u518a\u300d\u52a0\u4ee5\u8fa8\u8a8d\uff0c\u53ea\u8981\u5e8f\u5217\u76f8\u540c\uff0c\u4fbf\u6703\u7acb\u523b\u88ab\u5206\u89e3\u6389\uff0c\u90a3\u6bb5RNA\u6240\u4ee3\u8868\u7684\u57fa\u56e0\uff0c\u4e5f\u5c31\u7121\u5f9e\u8868\u73fe\u8d77\u4e86\u3002<\/p>\n<p>\u800c\u4eba\u985e\u5229\u7528\u9019\u500b\u6a5f\u5236\uff0c\u5c07\u6211\u5011\u60f3\u8981\u7814\u7a76\u7684\u57fa\u56e0\uff0c\u4ee5siRNA\u7684\u6280\u8853\u4f7f\u5b83\u4e0d\u8868\u73fe\uff0c\u89c0\u5bdf\u8a72\u751f\u7269\u6709\u4ec0\u9ebc\u6027\u72c0\uff0c\u4e5f\u65e9\u5df2\u4e0d\u662f\u65b0\u9bae\u4e8b\u4e86\u3002\u4f46\u662f\uff0c\u8981\u628a\u9019\u500b\u6280\u8853\u7528\u5728\u6bba\u87f2\u4e0a\uff0c\u666e\u6717\u514b\u7814\u7a76\u6240\u7684\u5718\u968a\uff0c\u9084\u771f\u662f\u50b7\u4e86\u4e00\u756a\u8166\u7b4b\u3002<\/p>\n<p>\u539f\u4f86\uff0c\u8981\u8b93\u96d9\u80a1RNA\u9032\u5165\u87f2\u7684\u9ad4\u5167\uff0c\u6700\u7c21\u4fbf\u7684\u65b9\u6cd5\u7576\u7136\u662f\u8b93\u87f2\u628a\u96d9\u80a1RNA\u81ea\u5df1\u5403\u4e0b\u53bb\uff1b\u6240\u4ee5\uff0c\u5982\u679c\u80fd\u5920\u8b93\u99ac\u9234\u85af\u8868\u73fe\u9019\u6bb5\u96d9\u80a1RNA\uff0c\u90a3\u9ebc\u87f2\u5152\u5011\u7528\u9910\u6642\u4fbf\u4e00\u4f75\u628a\u6bd2\u85e5\u7d66\u5403\u4e0b\u809a\u53bb\uff0c\u8c48\u4e0d\u7f8e\u54c9\uff1f<\/p>\n<p>\u4f46\u662f\u9019\u4ef6\u4e8b\u8aaa\u4f86\u5bb9\u6613\u4f5c\u4f86\u96e3\u3002\u9996\u5148\uff0c\u96d9\u80a1RNA\u901a\u904e\u87f2\u5152\u7684\u4e2d\u8178(midgut)\u6642\uff0c\u5982\u679c\u5168\u9ad4\u99ac\u4e0a\u5c31\u55da\u547c\u54c0\u54c9\u4e86\uff0c\u90a3\u5c31\u4ec0\u9ebc\u90fd\u4e0d\u7528\u60f3\u4e86\uff1b\u9084\u597d\u96d9\u80a1RNA\u7684\u78ba\u53ef\u4ee5\u7d93\u7531\u4e2d\u8178\u5438\u6536\uff0c\u4e26\u4e0d\u6703\u5168\u90e8\u9663\u4ea1\u3002\u63a5\u8457\uff0c\u8981\u8b93\u690d\u7269\u53ef\u4ee5\u8868\u9054\u96d9\u80a1RNA\uff1b\u904e\u53bb\u5728\u7389\u7c73\u7684\u5be6\u9a57\u4e2d\u5b8c\u5168\u6c92\u6709\u554f\u984c\uff0c\u4f46\u662f\u5230\u4e86\u99ac\u9234\u85af\u537b\u8e22\u5230\u4e86\u9435\u677f\u3002\u96d6\u7136\u8868\u73fe\u96d9\u80a1RNA\u7684\u8f49\u6b96\u57fa\u56e0\u7684\u78ba\u9032\u5165\u4e86\u99ac\u9234\u85af\uff0c\u4f46\u662f\u99ac\u9234\u85af\u7d30\u80de\u5167\u7684Dicer\u537b\u99ac\u4e0a\u8a8d\u51fa\u4e86\u5b83\uff0c\u4e26\u5c07\u5b83\u7684\u8868\u73fe\u7d66\u95dc\u6389\u4e86\u3002<\/p>\n<p>\u600e\u9ebc\u8fa6\u5462\uff1f\u7814\u7a76\u5718\u968a\u60f3\u5230\uff0c\u5982\u679c\u80fd\u5728\u8449\u7da0\u9ad4\u88e1\u9762\u8868\u73fe\u9019\u6bb5\u96d9\u80a1RNA\uff0c\u61c9\u8a72\u5c31\u4e0d\u6703\u6709\u554f\u984c\u3002\u70ba\u4ec0\u9ebc\u5462\uff1f\u56e0\u70ba\u8449\u7da0\u9ad4\u6e90\u81ea\u65bc\u85cd\u7da0\u83cc(cyanobacteria)\u9019\u7a2e\u539f\u6838\u751f\u7269\uff0c\u800c\u539f\u6838\u751f\u7269\u4e0d\u5177\u6709\u8fa8\u8b58\u96d9\u80a1RNA\u7684\u6a5f\u5236\u3002<\/p>\n<p>\u56e0\u6b64\uff0c\u7814\u7a76\u5718\u968a\u4fbf\u5c07\u53ef\u4ee5\u540c\u6642\u6216\u500b\u5225\u8868\u73fe\u5169\u500b\u57fa\u56e0\u7684\u96d9\u80a1RNA\u7684\u8f49\u6b96\u57fa\u56e0\uff0c\u8f49\u5165\u99ac\u9234\u85af\u7684\u8449\u7da0\u9ad4\u4e2d\uff1b\u9019\u5169\u500b\u57fa\u56e0\u90fd\u662f\u6240\u8b02\u7684\u5fc5\u9700\u57fa\u56e0(essential gene)\uff0c\u5982\u679c\u5b83\u5011\u4e0d\u8868\u73fe\uff0c\u87f2\u5152\u4fbf\u6703\u6b7b\u6389\u3002\u5176\u4e2d\u4e00\u500b\u662f\u7d30\u80de\u9aa8\u67b6(cytoskeleton)\u7684\u03b2-\u808c\u52d5\u86cb\u767d(\u03b2-actin)\uff0c\u53e6\u4e00\u500b\u5247\u662f\u8207\u6db2\u6ce1\u8f38\u9001\u76f8\u95dc\u7684Shrub\u57fa\u56e0(\u53e6\u540dVps32\u6216Snf7)\u3002\u6700\u5f8c\uff0c\u7814\u7a76\u5718\u968a\u767c\u73fe\uff0c\u55ae\u7368\u5e72\u64fe\u03b2-\u808c\u52d5\u86cb\u767d\u7684\u690d\u7269\uff0c\u6bba\u87f2\u7684\u6548\u679c\u975e\u5e38\u597d\uff0c\u5728\u7b2c\u4e94\u5929\uff0c\u4e00\u9f61\u5e7c\u87f2\u5c31\u5168\u90e8\u90fd\u6b7b\u5149\u4e86\uff1b\u800c\u5e72\u64feShrub\u57fa\u56e0\u3001\u6216\u662f\u540c\u6642\u5e72\u64fe\u03b2-\u808c\u52d5\u86cb\u767d\u8207Shrub\u57fa\u56e0\u7684\u690d\u7269\uff0c\u6bba\u87f2\u7684\u6548\u679c\u53cd\u800c\u4e0d\u7406\u60f3\uff0c\u540c\u6642\u5e72\u64fe\u5169\u500b\u57fa\u56e0\u7684\u690d\u7269\uff0c\u6bba\u87f2\u7684\u6548\u679c\u6bd4\u5e72\u64feShrub\u57fa\u56e0\u7684\u66f4\u5dee\uff1b\u611f\u89ba\u4e0a\uff0c\u5e72\u64feRNA\u7528\u5728\u6bba\u87f2\u4e0a\uff0c\u4f3c\u4e4e\u4e26\u4e0d\u662f\u591a\u591a\u76ca\u5584\u5462\uff01<\/p>\n<p>\u96d6\u7136\u9019\u500b\u767c\u73fe\uff0c\u4f3c\u4e4e\u63d0\u4f9b\u4e86\u672a\u4f86\u767c\u5c55\u6bba\u87f2\u5291\u7684\u53e6\u4e00\u689d\u8def\uff1b\u4f46\u662f\u4ee5\u79d1\u7f85\u62c9\u591a\u91d1\u82b1\u87f2\u9019\u6a23\u5584\u65bc\u6f14\u5316\u51fa\u6297\u85e5\u6027\u7684\u87f2\u5152\u4f86\u8aaa\uff0c\u7a76\u7adf\u9019\u500b\u65b9\u6cd5\uff0c\u6703\u4e0d\u6703\u8b93\u79d1\u7f85\u62c9\u591a\u91d1\u82b1\u87f2\u518d\u6b21\u7372\u53d6\u53e6\u4e00\u7a2e\u65b0\u80fd\u529b\u5462\uff1f\u9019\u7a2e\u80fd\u529b\uff0c\u662f\u5426\u6703\u4f7f\u7260\u5011\u6539\u8b8a\u03b2-\u808c\u52d5\u86cb\u767d\u7684\u5e8f\u5217\u5462\uff1f\u53ea\u80fd\u8aaa\uff0c\u751f\u7269\u7684\u4e16\u754c\uff0c\u6c38\u9060\u90fd\u6703\u8b93\u6211\u5011\u9a5a\u5947\uff0c\u6240\u4ee5\u300c\u5c31\u8b93\u6211\u5011\u770b\u4e0b\u53bb\u300d\uff5e<\/p>\n<p><strong>\u53c3\u8003\u6587\u737b\uff1a<\/strong>
